And yet, out of the princesses, only Luna and Celestia refer to each other as “sister”. And so does the rest of the cast. Additionally, I never heard Twilight or Cadence refer to Luna or Celestia as such.
 
In Slice of Life I felt like their bickering was more like an old married couple than sisters considering how I hear the way my Mom and Aunt argue and the tone is vastly different.
 
Old couple. Exactly what they are. They are both very old, and from what the episodes showed, can be quite quirky.
 
And speaking of the conversation from Slice of Life, I remember having a few conversations like that with my brother. Usually comes with miscommunication or failed expectations when visiting someone.

Honestly at this point with Cadence and Twilight ascending to Alicornhood it wouldn’t surprise me if they weren’t actually related but the “sister” moniker is used more for presenting a united front for the royal “family.”
 
Kind of like how familial terms are used in Japan among non-related younger/older children.
