Definitely specisist but not malicious. Interspecies relationships on a micro and macro level are far from the norm. Unicorns appear to be an upperclass so their relationship with other species is utilitarian business ventures. Pegasi and griffins get along but the stereotypical griffin attitude precludes frequent interaction. Earth ponies, besides cattle (sentient species at that) and the buffalo don’t really interact with any other races. They may see segregation as the norm and not think it worth fighting against.

Nah, I think she’s being pretty specisist here. We’re overlooking the impact Spike’s life had on ponies. Before he was hatched, ponies had never seen a dragon up close before except while fighting for their lives. Limestone’s unkind attitude was probably commonplace at the time.

We can possibly infer from Limestones’ attitude that she isn’t being speciesist towards the relationship of Twilight and Spike in that she is insulting their stance, she offers (somewhat misguided) points on how family actually operates. Dragons do not hold family to the same standards as Ponies do, but she doesn’t know that.
