Nope, they’re the same exact site, just alternative URLs. derpibooru.org is the site’s “default” URL, but you can use either one without issue.
However, cookies are based on which URL you’re using. So if you log into derpibooru.org, you won’t be logged into derpiboo.ru as well, unless you log in there separately.
