Yeah, but “sister-in-law” is the official term for it.
A sisterly bond doesn’t require blood, and many with a blood bond don’t see each other as sisters. That’s just how things go.

I think that’s the point. I assumed this pic showed them at around that age - Luna looks to be around her mid teens, and Celly appears to be in her early to mid 20s.

Yep, that’s it. I’m surprised there isn’t a tag for it already.
Because to me it looks like Cadance is genuinely surprised to be getting hugged by Twilight, and Twilight seems genuinely content. Some might interpret that to suggest that this happens rarely, either because it’s simply not often possible, or because Twilight hasn’t been the type of engage in physical contact with others.
Either was, whatever the reason, it’s pretty sad.
