In Lesson Zero towards the end Celestia was talking to Twilight about Spike. She called him “your friend Spike.” This shows that Spike is connected to Celestia through Twilight. There is a bit of distance in that relationship. I doubt she’s his adoptive mother.

Faust claims a lot of things, but half those things never show up in the show.
 
Twilight treats Spike more often than not like a mere slave. If Celestia was his adopted mom, he’d be visiting Canterlot far more often.

Classically, she has put the needs of her country ahead of her family, and she is endlessly busy even with Luna back… What makes you think she could care for a child?
