I just figured it out:
 
Fill both of her bars as far as you can, when the transparent bar is flashing, you have to turn the pump buttons to off then CONSTANTLY switch between fast and slow for the vibrator. It’ll fill VERY slowly, but it DOES get the job done.

Actually, that is not quite true. You don’t need to use specific characters to unlock the others, with the exception of Twi. The unlocks are as follows:  
Applejack - Use only the pump.  
Pinkie Pie - Use only the vibrator.  
Rarity - Overfill the tank.  
Fluttershy - Do not use the fast settings.  
Twilight - Achieve orgasm with the rest of the mane 6.  
Derpy - Achieve orgasm, but cum nothing (this will show up as a broken heart in the speech bubble).

How to unlock all characters: (Use - Settings - Unlocks)  
1: Rainbow Dash - Pump: Off Vibrator: Fast - (Unlocks Pinkie)  
2: Pinkie Pie - Pump: Fast Vibrator: Off - (Unlocks Applejack, Derpy)  
3: Applejack - Pump: Medium Vibrator: Slow - (Unlocks Fluttershy)  
4: Pinkie Pie - Pump: Medium Vibrator: Fast - (Unlocks Rarity)  
5: Rarity - Pump: Fast Vibrator: Fast - (Unlocks None)  
6: Fluttershy - Pump: Fast Vibrator: Fast - (Unlocks Twilight)

In order to overfill a tank, get the “pleasure” bar as close to the top as possible without her coming. The “milk” bar will rise faster when they’re close to climax, so you have to keep the pleasure bar set high while waiting for the “milk” to rise.

Does anyone know how to freaking overfill rainbow dash?
