Sounds like a good premis for One Night at Pinkie’s. Maybe someone will actually make it eventually.

I thought that Pinkie would be a good choice for the role of Flumpty becuase of how his immunity to the plot is similar to Pinkie’s fourth wall breaking abilities, you can see Pinkie using her fourth wall breaking abilities to randomly move to room to room.  
I can see Gummy playing the role of the Beaver as Gummy would blankly stare at the cameras when you check on him.  
The CMCs could be the Birthday Boy Blam, they would look harmless and cute as they appears in different room wearing cute costumes but when they get near their cute demeanor will be replace with a sadistic one especially when they’re standing at the hall door with the lights off.  
And the rest of main of MLP can start at their rooms doing things that you would normally see them do, like Twilight is in the library reading and Rarity in another room sewing(but if you looks hard you can find some disturbing things like Rarity is working a corpse) but when they leave their rooms their appearance will look different as they hunt you down.  
And throw Celestia and Luna with some MLP villains like Sombra and Chrysalis, Tirek in later nights. And I can see the game’s story being something like Pinkie would kidnap the protagonist and force him or her to play her game because she was bore and if the protagonist wins her game the protagonist will become her new friend but losing the game the protagonist will wind-up as cupcakes. As nights goes by Pinkie will invites some of her friends to help her play game each being premised to do whatever they want with the body to whomever manage to catch the the protagonist first.

Indeed. When it comes to Flumpty, Pinkie does seem like the more suitable choice. Twilight can be the Owl, one of the CMC can be Birthday Boy Blam and maybe Cheese Sandwich could be Grunkfuss the Clown. I don’t know about the rest of Flumpty’s friends, though.

After seeing this and hearing the Flumpty’s Jam song I only can picture this part of the song:  
Welcome to Twily’s  
Keep those lights off  
When they’re all turned off  
Then you’re safe  
If the lights are on  
she will come find you  
And rip you  
A brand new face…  
But I think Pinkie fit the role of Flumpty more then Twilight.  
Someone really need to make a MlP version of the One Night of Flumpty’s games.
