Applejack might be a dressmaker in Manehattan, Pinkie might be married to Big Mac or stuck on the rock farm, Fluttershy would probably still be an animal caretaker owing to her early flying disability, Rainbow would probably still be trying for the Wonderbolts, and Twilight probably wouldn’t have passed Magic Kindergarten with the flying colors she did, although she’d most definitely have passed at the top of her class. Rarity might still be a country bumpkin. Spike, of course, wouldn’t have hatched, or only from some other experiment.
 
The biggest change would be Nightmare Moon pretty much winning in one go, since the Elements of Harmony would be inoperable, unless Princess Celestia had the ability to use them all at once on her own. Without her Bearers, she’d have to throw down some actual solar power, possibly vaporizing a large portion of the map in the process. It also depends on whose orders Cerberus follows. If he’s independent, Tartarus might be safe. If Nightmare Moon’s arrival screwed things up royally over there, Tirek might break loose along with who-knows-what-else. If the resulting chaos of Unending Night gets big enough, Chrysalis might take the chaos as a sign of weakness and begin her war on Equestria early, and I don’t think Shining Armor can hold a shield that long without Cadence. With the war and CHAOS, Discord would be set free from his statue, resulting in another Capitol of Chaos. Without Cadence and the Elements and Spike, King Sombra’s return would allow him to easily take over the Crystal Empire and use the Crystal Heart to bring about a second reign of terror.
 
And meanwhile, Trixie’s antics would result in awakening the Star Creatures. No biggie.
 
The griffons probably would end up extinct, with nothing to unify them. Or maybe they take a look at the chaos and realize that, idol or no idol, they’d need to unify under one leader or another. Yakyakistan might withdraw even further into isolationism to wall off their southern border from the anarchy that was once Equestria.
 
Lord Discordance of Chaos, Queen Chrysalis, King Sombra, Lord Tirek, and Nightmare Moon would either set up agreements to have tense “peace” with one another or just go to outright war. Lord Discord and Lord Tirek might act like old pals, given that Discordance wouldn’t be all “weak”, and having an empire of his own. King Sombra might try to renew the wars of a thousand years past, if he’s sane, or just enslave the Crystal Empire and stay isolationist. Queen Chrysalis would probably take a look around and be more or less content with officially taking over Canterlot and using changeling subterfuge to take over other lands one by one. Patience is key.
 
Zecora would still be a shaman of the Everfree, maybe a bit more warlike, and embracing the whole “evil enchantress” thing to deter would-be enemies from entering the Forest. I imagine poison joke concoctions and illusion dust would help a lot in that regard.
 
We’d end up with quite a Game of Thrones style world, with everyone either morally grey or a villain, and everyone backstabbing everyone else, and the mortal ponies bearing the brunt of the collateral damage as slaves and peons. Frankly, Starlight Glimmer would be of trivial importance in this new world, likely getting killed in either a rain of exploding chocolate milk or a stray death beam from Lord Tirek. This not even taking into account the fact that most of the lands would probably be experiencing enormous food shortages because sunlight isn’t a thing anymore.
